Effect of guanidine hydrochloride(cas 50-01-1) concentration on superoxide dismutase (SOD) solution

To investigate the activity and conformational changes of superoxide dismutase (SOD) solution under different influencing factors.

Methods: The activity of SOD solution was determined by pyrogallol autoxidation rate method. The changes of SOD and tertiary structure with different influencing factors (pH value, ultrasonic and denaturant guanidine hydrochloride(cas 50-01-1)) were analyzed by steady-state fluorescence spectroscopy. The pH value decreased linearly with the activity of SOD, and the maximum emission wavelength of the fluorescence was shifted from 336 nm to blue with the decrease of pH value in the range of pH 2.2 ~ pH 7.0. The activity of SOD decreased with the increase of the number of ultrasonic waves, and the activity of SOD decreased with the increase of the concentration of guanidine hydrochloride(cas 50-01-1). When the concentration of guanidine hydrochloride reached 2.1 mol / L, the fluorescence intensity of SOD was the highest And the fluorescence intensity of the enzyme was higher than that of the natural enzyme after denaturation with guanidine hydrochloride.
