Guanidine hydrochloride(50-01-1) is a strong inhibitor of nuclease, it is not a strong enough denaturant, can allow complete RNA extracted from RNase-rich tissue. 

4-8M can be broken hydrogen bonds, there are two possible mechanisms: 1, denatured protein and guanidine hydrochloride, urea preferentially binds to form a denatured protein-denaturing agent complex, when the complex is removed, causing the ND reaction to move to the right. 2, guanidine hydrochloride, urea on the role of amino acid solubilization, can form hydrogen bonds, when the concentration is high, can destroy the water hydrogen bonds (the concentration of denaturant), the natural state of the protein into a complex, and ultimately lead to complete denaturation of the protein. 

Results of guanidine hydrochloride, urea is known as non-polar residues of the better solvent, so that the hydrophobic residues within the protein and the solubility of enhanced, guanidine hydrochloride, urea-induced degeneration is often irreversible.

High concentrations of urea denature the protein and inhibit Rnase activity.
