The addition of guanidine hydrochloride(cas 50-01-1) reduced the adsorption of BSA on PEG-600 surface

In this study, bovine serum albumin(BSA) was used as the object of study. The concentration of ammonium sulfate ((NH4) 2SO4) was 2.1mol·L-1 at 25 ℃, and the concentration of guanidine hydrochloride(GuHCl, cas 50-01-1) and the concentration of guanidine hydrochloride was 1.8 mol·L-1. Under different ammonium sulfate concentration, the enthalpy of adsorption of BSA in the adsorption of highly efficient hydrophobic interaction chromatographic packing (PEG-600) (SDT-A) and its thermodynamics, the thermodynamic functions ΔH, ΔS, ΔG and their components of BSA adsorbed on the surface of PEG-600 were obtained by the adsorption isotherm under the corresponding conditions. Scanning calorimetry(DSC) was used to determine the thermal stability of adsorbed BSA. Fourier transform infrared spectroscopy (FTIR) was used to determine the content of secondary structure of adsorbed BSA. The effects of guanidine hydrochloride and ammonium sulfate on the modified hydrophobic surface and to provide the theoretical basis for finding the best folding path of the denatured protein on the hydrophobic surface. 
The results of adsorption isotherm showed that the addition of guanidine hydrochloride(cas 50-01-1) reduced the adsorption capacity of BSA on PEG-600, while the addition of ammonium sulfate increased the adsorption capacity.
