The effect of guanidine hydrochloride(cas 50-01-1) on the endogenous fluorescence spectrum of human placental alkaline phosphatase was observed

The effect of guanidine hydrochloride(cas 50-01-1) on the endogenous fluorescence spectrum of human placental alkaline phosphatase was observed.

The results showed that when the concentration of guanidine was less than 1.0mol /L, the intensity of endogenous fluorescence emission decreased with the increase of guanidine concentration, and the fluorescence emission intensity increased, and when the guanidine concentration was more than 1.0mol /L. The maximum emission peak of the spectrum showed a red shift, and the red shift was stopped when the guanidine concentration was 4.0 mol /L. Suggesting that low concentrations of guanidine only perturbed the active site of the enzyme, while the high concentration of guanidine changed the overall conformation of the enzyme.
