The aqueous solution of lysozyme denatured with different concentrations of guanidine hydrochloride(cas 50-01-1) was scanned

In this study, two-beam ultraviolet spectrophotometer and circular dichroic CD spectrometer were used to scan the lysozyme aqueous solution of different concentrations of guanidine hydrochloride(cas 50-01-1) to study the changes of secondary and tertiary structure of lysozyme at different degree of denaturation. 

The results showed that with the concentration of guanidine hydrochloride(cas 50-01-1) from 0.4 to 2.6mol / L (protein concentration of 0.4mg / mL), the degree of degeneration of lysozyme also continue to strengthen, gradually lose its secondary and tertiary structure. But found that lysozyme were not completely denatured, still retain the same level of secondary and tertiary structure. At 25 ± 0.001 ℃ and 35 ± 0.001 ℃, the total enthalpy change (△ Hi) of the replacement of the lysozyme with different guanidine hydrochloride(cas 50-01-1) denatured on the surface of the hydrophobic solid (end PEG-600) hot method.
