Extraction of cord blood DNA by modified guanidine hydrochloride(cas 50-01-1) method for HLA genotyping

In order to compare the effects of classical guanidine hydrochloride(cas 50-01-1) extraction method and modified guanidine hydrochloride extraction method on the extraction of umbilical cord blood DNA, the two methods were studied in the umbilical cord blood phase compatibility antigen (HLA) genotyping. The DNA concentration and purity of 72 cord blood samples were extracted by DNA extraction with guanidine hydrochloride and modified guanidine hydrochloride extraction method. The results showed that the DNA concentration and purity were determined by sequence-specific primer polymerase reaction method (PCR-SSP ) compare the DNA extracted by these two methods. The results showed that the extraction of DNA from umbilical cord blood was successful. According to the quality monitoring of DNA, it was found that the DNA extracted with modified guanidine hydrochloride was superior to guanidine hydrochloride(cas 50-01-1) in quality. For HLA genotyping, DNA extraction can reduce the appearance of non-specific bands. Conclusion: The modified guanidine hydrochloride extraction method is not only simple and costly, but also can extract high purity DNA and reduce the appearance of non - specific bands in the least amount of blood. This method can fully meet the requirements of DNA extraction of cord blood samples and cord blood HLA genotyping.
