MES(4432-31-9) activates carboxyl beads act as activation buffer

A new type of bio-magnetic beads, which can enrich the H5N1 subtype avian influenza virus from large volume and low concentration virus liquid samples, was prepared. The effect of amino bead and carboxyl bead as biomagnetic bead carrier was compared. The results showed that activation of carboxy bead with pH 4.5 MES(4432-31-9) as activation buffer could bind 9.8 μg of fetuin, pH 4.5 PBS (phosphate buffer) to activation. The buffer can be combined with 9.25μg of fetal globulin; 5% glutaraldehyde as activator activated amino beads 1h more than 2.5%, 7.5% glutaraldehyde as activator per milliliter combined with fetal globulin, for 8.12 μg. The carboxyl-based bio-magnetic beads and the amino-type bio-magnetic beads can enrich the H5N1 subtype avian influenza virus, and the carboxyl-type bio-magnetic beads can be enriched to detect 15 times the virus dilution, the sensitivity is stronger than the amino-magnetic beads.
