HEPES(cas 7365-45-9) inhibits extracellular acidification to significantly inhibit cell killing

Cytotoxicity test showed that CO2 pneumoperitoneum had a significant killing effect on COLO 205 cells. Flow cytometry and cell morphology showed that induction of apoptosis was the main mode of action. The pH value of the extracellular environment decreased from 7.4 to 6.7 after the action of CO2. The inhibition of extracellular acidification by HEPES(cas 7365-45-9) could significantly inhibit cell killing.

To study the killing effect of hot CO2 pneumoperitoneum on colon cancer cells in vitro and to explore its possibility as a new method of peritoneal metastasis of colorectal cancer. 

Methods: The acute lung pneumoperitoneum model was established by W ST-8 method. The cytotoxicity of hot CO2 pneumoperitoneum (42 ℃ ~ 44 ℃, 2 ~ 4 h) on colon cancer cell line COLO 205 was detected by flow cytometry. Hoechst 33342 / PI fluorescence microscopy, transmission electron microscopy and other clear cell death mechanism; PT100 temperature and pH meter to detect the extracellular temperature and pH changes; extracellular acidification inhibition test to identify the role of extracellular acidification.

Conclusion: Hot CO2 pneumoperitoneum can kill colon cancer cells by inducing apoptosis. Cell killing is related to heat and CO2 acidification. Hot CO2 pneumoperitoneum has potential for peritoneal metastasis of colon cancer.
