Cr (Ⅵ) - GSH intermediate complex was formed in HEPES(7365-45-9) buffer solution

To study the mechanism of interaction between potassium dichromate and reduced glutathione (GSH) and DNA to understand the carcinogenesis of Cr (Ⅵ) in vivo.

Methods: 1: 10 K2Cr2O7 and GSH solution were used to form Cr (Ⅵ) -GSH intermediate complex, and a DNA solution of 1.4 × 10-3 mol / L was prepared. Brominated ethidium bromide (EB) was used as DNA fluorescent probe, The effects of Cr (Ⅵ) -GSH complex on DNA conformational changes were studied by UV spectroscopy, fluorescence spectroscopy, scatchard and DNA thermal denaturation.

Results: Cr (Ⅵ) - GSH intermediate complex was formed by reaction of Cr (Ⅵ) with GSH in HEPES(7365-45-9) buffer solution with pH = 7.4. The complexes do not interact with DNA, nor do they interact with the DNA phosphoric acid skeleton, but instead act on the DNA base site, destroying the DNA secondary structure. The complexes of the intermediate complexes are unstable and the final product Cr (Ⅲ ) can be cross-linked with DNA, the whole process about 1 h to complete.

CONCLUSION: Cr (Ⅴ) complex formed by the reaction of potassium dichromate and reduced glutathione (GSH) plays an important role in the induction of DNA denaturation.
